High resolution insights into binding and folding by direct detection of Hydrogen bonds. A review of "Temperature-dependence of Protein Hydrogen Bond Properties as Studied by High-resolution NMR" Cordier et al. JMB 2002, 317, 739-752.

and

"Ligand-Induced Strain in Hydrogen Bonds of the c-Src SH3 Domain Detected by NMR" Cordier et al. JMB 2000, 304, 497-505.

Trans-hydrogen bond amide nitrogen to carbonyl carbon (h3JNC') coupling constants provide a means for the direct detection of changes in hydrogen bond length and strength. The length of a H-bond is exponentially related to the magnitude of the h3JNC' coupling constant. These coupling constants have been used to study folding and ligand binding of proteins. This property enables the study of conformational changes at picometer resolution.
Building
